Overview

Nivolumab + vorolanib is an investigational combination therapy consisting of nivolumab, a monoclonal antibody targeting the programmed cell death protein 1 (PD-1), and vorolanib, an oral small-molecule multi-kinase inhibitor. Nivolumab acts as an immune checkpoint inhibitor by blocking PD-1 on T cells, thereby enhancing anti-tumor immune responses. Vorolanib inhibits multiple receptor tyrosine kinases involved in angiogenesis and tumor growth, including vascular endothelial growth factor receptors (VEGFR), platelet-derived growth factor receptors (PDGFR), colony-stimulating factor 1 receptor (CSF1R), c-Kit, and FMS-like tyrosine kinase 3 (FLT3). The rationale for combining these agents is to overcome resistance to immunotherapy by modulating the tumor microenvironment through both immune activation and inhibition of angiogenesis pathways. This combination has been evaluated in phase 1/2 clinical trials for patients with refractory thoracic malignancies such as non-small cell lung cancer (NSCLC) and thymic carcinoma[1][2][4][6].
