NIX1-armed oncolytic herpes simplex virus is an experimental oncolytic immunotherapy designed to treat primary brain tumors, such as glioblastoma (GBM). It consists of an oncolytic herpes simplex virus (oHSV) engineered to express NIX1, a peptide that interferes with the binding of NOTCH ligands to their receptors. By blocking the NOTCH signaling pathway—a pathway often exploited by GBM stem cells to resist treatment and recruit myeloid-derived suppressor cells (MDSCs)—this therapy aims to suppress tumor regrowth and enhance immune-mediated tumor killing. Preclinical studies in intracranial tumor models have demonstrated that NIX1-armed oHSV can significantly increase therapeutic benefit and induce macrophage migration into the tumor microenvironment.
