NIX2-armed oncolytic herpes simplex virus is an experimental oncolytic immunotherapy designed to treat primary brain tumors, specifically glioblastoma (GBM). It consists of an oncolytic herpes simplex virus (oHSV) engineered to express NIX2, a peptide that blocks the NOTCH signaling pathway by interfering with the binding of NOTCH ligands to their receptors. NOTCH signaling is often exploited by glioblastoma stem cells to resist treatment and promote tumor regrowth. By combining the direct oncolytic activity of the virus with NIX2-mediated NOTCH blockade, the therapy aims to suppress tumor growth, reduce the recruitment of immunosuppressive monocytic myeloid-derived suppressor cells (MDSCs), and enhance the anti-tumor immune response. Preclinical studies in intracranial tumor models have demonstrated significant therapeutic benefits and increased macrophage migration into the tumor microenvironment.
