NJ3d is a synthetic phenothiazine derivative developed as a dual-target inhibitor for the treatment of Alzheimer's disease. It is designed to simultaneously inhibit Monoamine Oxidase B (MAO-B) and Acetylcholinesterase (AChE). By targeting AChE, it aims to increase acetylcholine levels to improve cholinergic neurotransmission, while its inhibition of MAO-B is intended to reduce oxidative stress and neuroinflammation associated with the degradation of biogenic amines. In preclinical studies, NJ3d demonstrated high binding affinity in molecular docking simulations and showed potential in cytotoxicity evaluations, although its blood-brain barrier (BBB) permeability was characterized as mediocre, suggesting a need for further optimization or nanoformulation delivery strategies.
