**NJA-730** is a novel immunosuppressive therapeutic developed by NapaJen Pharma, consisting of an anti-CD40 oligonucleotide (specifically a small interfering RNA or siRNA targeting CD40) complexed with a proprietary beta-glucan (schizophyllan) delivery vehicle that selectively targets Dectin-1-expressing dendritic cells and macrophages. It inhibits CD40 expression to block the CD40-CD40L costimulatory pathway, preventing T-cell activation and immune responses. NJA-730 demonstrated safety and tolerability in a Phase 1 trial in healthy volunteers, with a short half-life of ~30 minutes, linear pharmacokinetics, and no serious adverse events; preclinical mouse models showed efficacy in prolonging bone marrow engraftment and preventing acute graft-versus-host disease (aGVHD).[1][2][3]
