NJK14047 is a potent and selective small molecule inhibitor of p38 mitogen-activated protein kinase (p38 MAPK), specifically targeting the p38α isoform. It was originally developed as a potential treatment for inflammatory lung conditions such as influenza, but subsequent research has demonstrated its antiviral and anti-inflammatory effects in various disease models. Mechanistically, NJK14047 inhibits the differentiation of naïve CD4+ T cells into Th1, Th2, and Th17 subsets and suppresses proinflammatory cytokine production. Preclinical studies have shown efficacy in models of atopic dermatitis, rheumatoid arthritis, psoriasis, hepatitis B virus infection, and other inflammation-driven diseases by reducing inflammatory cytokines and viral replication[1][3][6][7][9].
