NK-92-αFR-28BBζ is a third-generation **chimeric antigen receptor (CAR)-engineered NK-92 cell therapy** designed to target and kill cancer cells that overexpress **folate receptor alpha (αFR)**, notably in ovarian cancer. The CAR construct is fully humanized and built on a single-chain variable fragment (scFv) derived from the human antibody C4. It combines three intracellular domains: **CD28, CD137 (4-1BB), and CD3ζ**, which together enhance cytotoxic activation, proliferation, cytokine release (especially IFN-γ and TNF-α), and persistence while reducing apoptosis compared to earlier generation αFR-targeting CAR-NK therapies. Engineered NK-92-αFR-28BBζ cells demonstrate high antigen-specific cytotoxicity in vitro and significant tumor regression and survival benefit in ovarian cancer xenograft models. This cell therapy is under preclinical and early clinical investigation as a targeted immunotherapy for αFR-positive tumors, especially ovarian cancer[1][3].
