NK cell-enriched donor lymphocyte infusion (NK-DLI) + DUK-CPG-001 is a combination investigational therapy used in the context of allogeneic stem cell transplantation for hematologic malignancies. NK-DLI refers to infusions of donor-derived, natural killer (NK) cells that are enriched and administered to patients post-transplant to enhance graft-versus-leukemia effects. DUK-CPG-001 is a synthetic CpG-rich oligonucleotide that acts as an agonist of Toll-like receptor 9 (TLR9), stimulating innate immune responses by activating intracellular TLR9 and promoting immunopotentiation[1][2][3]. The combination aims to improve anti-tumor immunity by both direct cytotoxicity from NK cells and immune activation via TLR9 stimulation. This approach has been studied in phase 2 clinical trials primarily at Duke University for patients with myeloid or lymphoid malignancies following reduced intensity or non-myeloablative allogeneic stem cell transplantation[2][5].
