Overview

NK cell infusion (University of Wisconsin, Madison) is an investigational adoptive cell therapy consisting of donor-derived natural killer (NK) cells. Developed for use in pediatric patients undergoing reduced-intensity conditioning (RIC) haploidentical hematopoietic stem cell transplantation (HSCT), the therapy involves the infusion of NK cells selected from a haploidentical donor (typically a parent). The cells are processed using the CliniMACS cell separation system to deplete T-cells, thereby reducing the risk of graft-versus-host disease (GVHD) while retaining the potent anti-tumor effector functions of NK cells. These cells target malignant cells through various activating receptors, such as NKG2D and natural cytotoxicity receptors (NCRs), and are being evaluated for the treatment of recurrent or refractory acute lymphoblastic leukemia (ALL), acute myeloid leukemia (AML), and high-risk solid tumors including pediatric sarcomas.
