Drug intelligence / Profile preview
NK012 is a polymeric micelle formulation designed to deliver SN-38, the active metabolite of irinotecan (CPT-11), which is a potent DNA topoisomerase I inhibitor. In this nanodevice formulation, SN-38 is covalently attached to the block copolymer PEG-PGlu and self-assembles in aqueous media to form water-soluble micelles. This design increases the solubility and tumor-targeting ability of SN-38 compared to conventional formulations. Unlike irinotecan, which requires metabolic activation by enzymes to release SN-38, NK012 releases its active ingredient via hydrolysis directly at the tumor site. The mechanism of action involves inhibition of DNA topoisomerase I, leading to DNA breaks and apoptosis in cancer cells. Preclinical studies have shown that NK012 accumulates at high concentrations in tumors and may cause less severe gastrointestinal toxicity than irinotecan. Developed primarily by Nippon Kayaku for oncology indications such as triple-negative breast cancer, small cell lung cancer, colorectal cancer, and multiple myeloma, it has reached phase II clinical trials for several solid tumors[1][2][3][4].
