NK012 + carboplatin is a combination therapy under investigation for the treatment of solid tumors. NK012 is a polymeric micelle formulation containing SN-38, the active metabolite of irinotecan (CPT-11), which acts as a topoisomerase I inhibitor. Unlike CPT-11, NK012 releases SN-38 via hydrolysis and does not require metabolic conversion by enzymes. This allows for direct delivery of the active agent to tumor cells and may result in improved antitumor activity and reduced gastrointestinal toxicity compared to irinotecan[6]. Carboplatin is an organoplatinum alkylating agent that works by forming DNA crosslinks and monoadducts, thereby inhibiting DNA synthesis and transcription in cancer cells[1][4]. The combination aims to leverage both agents' mechanisms for enhanced efficacy against various solid tumors.
