NK121 is an experimental platinum-based antineoplastic agent developed by Nippon Kayaku. Chemically, it is a carboplatin analog, specifically identified as cis-1,1-cyclobutanedicarboxylato(2R)-2-methyl-1,4-butanediammineplatinum(II). Like other platinum complexes such as cisplatin and carboplatin, NK121 functions as a DNA alkylating agent that forms inter- and intra-strand cross-links, thereby inhibiting DNA replication and transcription and inducing apoptosis in rapidly dividing cancer cells. Research into NK121 has primarily focused on its synergistic effects when combined with hyperthermia, where it has demonstrated enhanced tumor growth delay in melanoma models. The compound was designed to offer a therapeutic profile with reduced nephrotoxicity compared to first-generation platinum drugs like cisplatin, although studies have noted that its toxicity profile can be altered when administered in conjunction with heat therapy.
