NK46141 is a monoclonal antibody developed by Nippon Kayaku that specifically targets single-chain type IV collagen (SCCOL-IV), an alternative gene product of the COL4A1 gene. SCCOL-IV is distinct from the standard triple-helical type IV collagen found in normal basement membranes and is specifically expressed in tumor tissues and cultured tumor cells. NK46141 binds to SCCOL-IV with high affinity and does not cross-react with triple-helical type IV collagen. In preclinical studies, prophylactic administration of NK46141 suppressed tumor growth in mouse models, likely by inhibiting tumor-associated vasculogenesis, as SCCOL-IV is localized in vessel lumens during tumor progression. It is being investigated as a potential therapeutic and diagnostic agent for various malignancies, including lung, liver, renal, and pancreatic cancers.
