Overview

NK521 is an adoptive natural killer (NK) cell therapy developed by Base Therapeutics for the treatment of advanced solid tumors and associated malignant ascites. NK cells are a critical component of the innate immune system, capable of recognizing and eliminating malignant cells through direct cytotoxicity and the secretion of pro-inflammatory cytokines without the need for prior antigen sensitization. NK521 is currently being evaluated in a Phase 1 clinical trial where it is administered via two distinct routes: intravenous infusion for patients with small-volume ascites and intraperitoneal perfusion for those with large-volume symptomatic ascites. This dual approach aims to address both systemic tumor burden and localized peritoneal carcinomatosis in patients with relapsed or refractory cancers, including liver, gastric, colorectal, and ovarian malignancies.
