NK92MI + HS20 is a genetically engineered combination cell therapy product consisting of NK92MI cells expressing a chimeric antigen receptor (CAR) based on the HS20 single-chain antibody fragment, which specifically targets glypican-3 (GPC3), a surface antigen highly expressed in hepatocellular carcinoma (HCC). The HS20 antibody recognizes the heparan sulfate chains present on GPC3. The CAR construct includes a human IgG1 hinge, CD28 transmembrane and intracellular domains, 4-1BB intracellular domain, and CD3zeta intracellular domain. The therapy is designed to enhance the cytotoxicity of natural killer (NK) cells against GPC3-positive tumor cells, aiming for targeted immunotherapy in HCC.
