**NK:BOB1-TCR + IL-15** is an investigational, engineered cell therapy consisting of natural killer (NK) cells that have been genetically modified to express a T cell receptor (TCR) specific for the BOB1 antigen (a B cell-specific transcription factor, also known as POU2AF1), combined with autonomous expression of interleukin-15 (IL-15). This combination endows the NK cells with dual mechanisms of cytotoxicity—TCR-mediated, directed against BOB1/HLA-B*07:02 positive tumor cells (such as multiple myeloma), and innate NK cell-mediated cytotoxicity, while IL-15 production supports the proliferation, survival, and persistence of the engineered cells without external cytokine support. Preclinical studies show enhanced anti-tumor activity and improved persistence in vivo, but also note the risk of potential toxicity due to cell accumulation in organs. The approach is being investigated for the treatment of B cell malignancies, especially multiple myeloma[1][4].
