NKAB-CLL1

NKAB-CLL1 (also referred to as NKAE-CLL1) is an investigational bispecific Natural Killer (NK) cell engager developed by NKarta for the treatment of myeloid malignancies, including acute myeloid leukemia (AML). The therapeutic is designed to bridge NK cells and tumor cells by simultaneously binding to CD16a (FcγRIIIa) on NK cells and CLL-1 (C-type lectin-like molecule-1, or CLEC12A) on leukemic blasts and leukemic stem cells. This dual binding facilitates the formation of an immunological synapse, leading to NK cell activation, degranulation, and the targeted destruction of CLL-1-expressing malignant cells. CLL-1 is a particularly attractive target in AML because it is highly expressed on the majority of AML cells but is absent on healthy hematopoietic stem cells, potentially offering a wider therapeutic window compared to other myeloid targets.
