NKD1-specific TCR-T cells are an investigational T cell receptor (TCR) engineered T cell therapy designed for the treatment of microsatellite stable (MSS) metastatic colorectal cancer (mCRC). The therapy utilizes naive CD8+ T cells engineered to express a high-affinity TCR specific for a peptide derived from the fetal WNT program gene NKD1 (Naked cuticle homolog 1). This peptide is presented by the HLA-A*02:01 (HLA-A2) molecule, which is the most common HLA allele. The target was identified through immunopeptidomic profiling of patient-derived organoids (PDOs), revealing that NKD1 is recurrently presented in mCRC and fetal tissues but is undetectable in healthy adult tissues. In preclinical studies, these engineered T cells demonstrated robust cytokine activation and potent, antigen-dependent cytotoxicity against mCRC cells with a favorable therapeutic index. The therapy is being developed by researchers at Memorial Sloan Kettering Cancer Center.
