Drug intelligence / Profile preview
An NKG2A blocking antibody is a type of monoclonal antibody designed to target and block the natural killer group 2 member A (NKG2A) receptor, an inhibitory checkpoint receptor expressed on natural killer (NK) cells and a subset of cytotoxic CD8+ T cells. NKG2A forms a heterodimer with CD94 and binds to its ligand, human leukocyte antigen E (HLA-E), which is frequently overexpressed on various tumor cells. This interaction transmits inhibitory signals that suppress the cytotoxic activity of NK and T cells, allowing tumors to evade immune surveillance. By blocking the NKG2A/CD94-HLA-E interaction, NKG2A blocking antibodies unleash the anti-tumor effector functions of these immune cells. Several NKG2A blocking antibodies, such as monalizumab, BMS-986315, and S095029, are being evaluated in clinical trials, often in combination with other immune checkpoint inhibitors (like PD-1/PD-L1 blockers) or tumor-targeting antibodies to enhance anti-cancer efficacy.
