NKG2C-KE is a trispecific killer engager biologic constructed to direct and activate natural killer (NK) cells expressing the NKG2C receptor, providing targeted delivery of interleukin-15 (IL-15) and redirecting these adaptive NK cells to CD33-positive tumor cells[1]. Its mechanism includes engaging NKG2C^+^ NK cells, delivering IL-15 specifically to this subpopulation, and linking these adaptive NK cells to tumor-associated antigens (specifically CD33), thereby boosting cytotoxicity and persistence of NK cells against acute myeloid leukemia (AML) cells[1]. Developed as an advanced immunotherapy tool, NKG2C-KE expands and activates NKG2C^+^ adaptive NK cells, mimicking signals seen in cytomegalovirus (CMV) infection, and offers a precision therapeutic approach for targeting AML by promoting proliferation, degranulation, cytokine production, and cytotoxic activity against CD33^+^ blasts and cell lines[1].
