NKG2D-ACE2 CAR-NK Cell is a universal, off-the-shelf chimeric antigen receptor (CAR) natural killer (NK) cell therapy engineered from cord blood. These cells are modified to express both the NKG2D and ACE2 proteins as part of their CAR construct, enabling them to target and eliminate virus-infected or malignant cells. The therapy also secretes an IL-15 superagonist to enhance NK cell activity and persistence, as well as a GM-CSF-neutralizing single-chain variable fragment (scFv) to reduce the risk of cytokine release syndrome. The dual targeting mechanism allows these cells to recognize SARS-CoV-2-infected cells via ACE2 binding to the viral spike protein and stress-induced ligands via NKG2D, making it a promising candidate for COVID-19 treatment. Additionally, its design aims for improved safety over traditional CAR-T therapies by minimizing severe immune-related side effects[1][2].
