NKG2D-BBz CAR-T is an autologous or allogeneic chimeric antigen receptor (CAR) T-cell therapy engineered to target cells expressing ligands for the Natural Killer Group 2 Member D (NKG2D) receptor. The CAR construct is composed of the extracellular domain of the human NKG2D receptor, which naturally binds to a variety of stress-induced ligands (NKG2DL) such as MICA, MICB, and ULBP1-6. These ligands are typically absent or expressed at low levels on healthy tissues but are frequently upregulated on the surface of malignant cells, including those in hepatocellular carcinoma (HCC). The intracellular signaling portion of the CAR includes the 4-1BB (CD137) costimulatory domain and the CD3ζ signaling domain, characterizing it as a second-generation CAR. Upon binding to NKG2D ligands on tumor cells, the CAR-T cells undergo activation, proliferation, and exert potent cytotoxic activity to eradicate the target cells. Preclinical studies have demonstrated significant efficacy in hepatocellular carcinoma models.
