NKG2D CAR mRNA-engineered NK cells are a form of cell therapy in which natural killer (NK) cells are genetically modified using messenger RNA (mRNA) to express a chimeric antigen receptor (CAR) that targets ligands for the activating receptor NKG2D. This engineering enhances the ability of NK cells to recognize and kill tumor cells expressing NKG2D ligands. The approach is being investigated as an immunotherapy for both hematological malignancies and solid tumors, with preclinical studies showing enhanced anti-tumor activity compared to conventional memory CAR-T cell therapies. Some constructs also coexpress cytokines such as IL-15 to further boost anti-tumor efficacy[1].
