NKG2d-CTL refers to a cell therapy product consisting of cytotoxic T lymphocytes (CTLs) that have been genetically engineered to express the NKG2D receptor, typically as part of a chimeric antigen receptor (CAR) construct. NKG2D is an activating receptor found on natural killer (NK) cells and some T cells, which recognizes stress-induced ligands such as MICA, MICB, and ULBP proteins that are upregulated on the surface of tumor or infected cells. By engineering CTLs to express NKG2D-CARs, these modified T cells gain the ability to recognize and kill cancerous or otherwise abnormal cells expressing NKG2D ligands. This approach is being investigated primarily for hematologic malignancies and solid tumors where these ligands are present. The mechanism involves direct cytotoxicity against target cells upon ligand recognition, secretion of cytokines, and potential modulation of the tumor microenvironment[1][7]. The therapy is in early clinical development.
