**NKp30-CD28 CAR T cells** are genetically engineered T cells that express a chimeric antigen receptor (CAR) based on the NKp30 natural cytotoxicity receptor, fused with the CD28 co-stimulatory domain and CD3ζ signaling domain. NKp30 is a receptor naturally expressed on NK cells, recognizing the B7-H6 ligand found on many tumor cells but rarely on normal cells. By combining NKp30’s extracellular domain with the intracellular signaling domains of CD28 and CD3ζ, these CAR T cells are designed to specifically target and kill B7-H6-expressing tumor cells, activating cytotoxicity and cytokine production (such as IFN-γ) in an MHC-independent manner. CD28 enhances T cell activation, survival, proliferation, and anti-tumor activity via PI3-kinase pathway activation. These cells have shown efficacy in preclinical models for killing various B7-H6-positive tumor cells and inducing anti-tumor immune memory[1][3].
