NKR-2 cells + FOLFOX is an investigational combination therapy under clinical development for cancer, primarily metastatic colorectal cancer. It combines **NKR-2 cells**, which are autologous T cells genetically engineered to express the NKG2D chimeric antigen receptor (CAR), with the standard chemotherapy regimen **FOLFOX**—a combination of folinic acid (leucovorin), fluorouracil (5-FU), and oxaliplatin. NKR-2 cells recognize a broad range of tumor-expressed ligands (MIC-A, MIC-B, ULBP1-6), enabling multi-targeted tumor cell killing, immune modulation (reducing regulatory T cells and myeloid-derived suppressor cells, recruiting macrophages), and anti-angiogenic activity. FOLFOX works by directly inhibiting DNA synthesis and cell division in rapidly dividing cancer cells. The combination aims to enhance anti-tumor efficacy by integrating the direct cytotoxic effects of chemotherapy with the immune-mediated and multi-pronged mechanisms of CAR T NKR-2 cells.
