Overview

NKS-5 (also known as NKS-05) is a selective 5-hydroxytryptamine receptor 4 (5-HT4) agonist originally developed by Nikken Chemicals (which later merged into Kowa Company). The compound was primarily investigated for its potential therapeutic application in cognitive disorders, specifically Alzheimer's disease and other forms of dementia. By activating 5-HT4 receptors in the central nervous system, NKS-5 stimulates the release of acetylcholine in the hippocampus and frontal cortex, which are critical areas for learning and memory. Furthermore, research indicated that NKS-5 could modulate the processing of amyloid precursor protein (APP) toward the non-amyloidogenic alpha-secretase pathway, potentially reducing the production of neurotoxic beta-amyloid. Although 5-HT4 agonists are frequently utilized for gastrointestinal motility disorders, NKS-5 was specifically targeted for its pro-cognitive and neuroprotective properties.
