NKTR-0165 is a novel, first-in-class bivalent monoclonal antibody that acts as an agonist of tumor necrosis factor receptor type II (TNFR2). It is designed to selectively stimulate TNFR2 activity without modulating TNFR1 signaling. This selective activation enhances the function and proliferation of regulatory T cells (Tregs), which are critical for maintaining immune tolerance and suppressing inflammation. By upregulating proteins essential for Treg stability and function, NKTR-0165 aims to reduce inflammation in autoimmune diseases. The drug was developed through a collaboration between Nektar Therapeutics and Biolojic Design, leveraging Biolojic’s proprietary technology platform. Preclinical studies have demonstrated efficacy in models of delayed-type hypersensitivity by enhancing Treg-mediated immunosuppression, with potential applications in diseases such as multiple sclerosis, ulcerative colitis, vitiligo, and other autoimmune disorders[1][2][3][4][5][7][9].
