NKTR-102 + cetuximab is a combination therapy under investigation for the treatment of solid tumors, particularly metastatic or locally advanced colorectal cancer. NKTR-102 (also known as etirinotecan pegol or PEG-irinotecan) is a PEGylated form of irinotecan, designed to be a long-acting topoisomerase I inhibitor that provides sustained release of its active metabolite SN38. Cetuximab is an epidermal growth factor receptor (EGFR) monoclonal antibody that inhibits EGFR signaling. The combination aims to leverage the cytotoxic effects of prolonged SN38 exposure from NKTR-102 with the targeted inhibition of EGFR by cetuximab. Clinical studies have shown manageable toxicity and evidence of antitumor activity in patients with refractory solid tumors and colorectal cancer[1][2][3][4][5].
