Overview

NKTR-105 is a novel PEGylated polymer-drug conjugate of the taxane chemotherapy agent docetaxel, developed by Nektar Therapeutics using its proprietary polymer conjugate technology. By attaching polyethylene glycol (PEG) chains to docetaxel, NKTR-105 is designed to optimize the pharmacokinetic profile of the parent drug, resulting in a prolonged half-life and enhanced accumulation within tumor tissues via the enhanced permeability and retention (EPR) effect. Like docetaxel, its mechanism of action involves binding to and stabilizing microtubules, thereby inhibiting cell division and inducing apoptosis. NKTR-105 was evaluated in Phase 1 clinical trials for the treatment of various advanced solid tumors, including breast, lung, and prostate cancers, with the goal of improving efficacy and reducing systemic toxicities such as neutropenia compared to standard docetaxel.
