Overview

NKTR-125 is an early-stage PEGylated version of recombinant human interleukin-2 (IL-2), developed by Nektar Therapeutics for the treatment of various cancers. It was designed using Nektar's proprietary polymer conjugation technology to extend the half-life of IL-2 and potentially improve its safety profile compared to high-dose aldesleukin (Proleukin). The mechanism involves binding to the IL-2 receptor complex to stimulate the proliferation and activation of effector T cells and natural killer (NK) cells. However, NKTR-125 was largely superseded in Nektar's development pipeline by NKTR-214 (bempegaldesleukin), a more optimized PEG-IL-2 prodrug designed for biased signaling toward the IL-2 receptor beta-gamma (IL-2Rβγ) complex.
