Overview

NKTR-255 is an investigational, long-acting, polymer-conjugated interleukin-15 (IL-15) receptor agonist designed to enhance the immune system’s natural ability to fight cancer. It mimics the biological activity of endogenous IL-15 and engages the full IL-15 receptor complex (IL‑15Rα/IL‑2Rβγ), leading to increased proliferation and survival of natural killer (NK) cells and memory CD8+ T cells. This results in enhanced antitumor immune responses and improved immunological memory. NKTR-255 is engineered using polyethylene glycol modification for extended half-life, overcoming rapid clearance issues seen with recombinant IL‑15 therapies. It has shown promise both as monotherapy and in combination with other agents such as CAR-T cell therapies, monoclonal antibodies (e.g., daratumumab, rituximab), or checkpoint inhibitors in hematologic malignancies and solid tumors[1][3][4][5][6].
