NKTR-255 + cetuximab is an experimental combination therapy comprised of NKTR-255, a polymer-conjugated interleukin-15 (IL-15) receptor agonist, and cetuximab, a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR). NKTR-255 is designed to stimulate the proliferation and survival of natural killer (NK) cells and CD8+ T cells by engaging the IL-15 receptor complex, potentially increasing the anti-tumor immune response and improving immunotherapy outcomes. Cetuximab binds EGFR, inhibiting signals necessary for tumor cell growth and survival, and enables immune-mediated mechanisms such as antibody-dependent cellular cytotoxicity. The combination aims to augment anti-tumor immunity through complementary mechanisms, with NKTR-255 enhancing immune effector cell number and function, while cetuximab blocks EGFR pathways and enables immune recognition of tumor cells. The combination is in clinical investigation for relapsed/refractory head and neck squamous cell carcinoma (HNSCC) and colorectal carcinoma (CRC)[1][4][7].
