NKTR-262 is a PEG-conjugated small molecule agonist of toll-like receptor 7 and 8 (TLR7/8), developed for intratumoral administration in cancer immunotherapy. The drug consists of a four-arm polyethylene glycol (PEG) molecule, each arm attached to a small-molecule TLR7/8 agonist via hydrolyzable glycine linkers. This design allows for sustained exposure within the tumor microenvironment while minimizing systemic exposure, thereby enhancing tolerability. NKTR-262 activates innate immunity by stimulating myeloid cells and promoting tumor antigen release and presentation, which leads to an immunostimulatory milieu that supports adaptive immune responses. It has been studied both as monotherapy and in combination with bempegaldesleukin (BEMPEG), an IL-2 pathway agonist, showing synergistic effects in preclinical models by expanding activated CD8+ T cells and improving antitumor efficacy[1][3][4][6][8].
