Overview

This is an investigational **combination immunotherapy** regimen comprising three agents: - **NKTR-262**: a small molecule agonist of toll-like receptors 7/8, administered intratumorally to activate antigen-presenting cells (APCs) and stimulate a local tumor immune response by activating dendritic cells and promoting the creation of new antigen-specific cytotoxic T cells. - **Bempegaldesleukin (NKTR-214)**: a CD122-preferential interleukin-2 (IL-2) pathway agonist engineered for systemic administration that selectively stimulates and expands proliferative CD8+ T cells and natural killer (NK) cells, ideally amplifying systemic anti-tumor immune response while minimizing toxicity. - **Nivolumab**: a monoclonal antibody immune checkpoint inhibitor targeting programmed cell death protein 1 (PD-1), relieving inhibitory signaling in T cells to further promote an anti-tumor immune response. The combination is under investigation for the treatment of a variety of solid tumors, most notably **melanoma**, as well as merkel cell carcinoma, triple negative breast cancer, renal cell carcinoma, colorectal cancer, head and neck squamous cell carcinoma, and sarcoma. Mechanistically, the combination is designed to couple local innate immune activation (via NKTR-262), systemic adaptive immune expansion (via bempegaldesleukin), and checkpoint inhibition (via nivolumab) to enhance anti-tumor efficacy[1][3][5][9].
