NKTR-288 is an investigational PEGylated interferon gamma (IFN-γ) agonist being developed by Nektar Therapeutics for the treatment of solid tumors. It is a polymer-conjugated interferon gamma mutein designed with site-specific conjugation to modify binding to the IFN-γ receptor and optimize the duration of signaling. The drug is engineered to have reduced affinity for both the IFN-γ receptor and heparin, which decreases target-mediated drug disposition and increases bioavailability compared to native interferon gamma. Preclinical studies have demonstrated that NKTR-288 can upregulate MHC Class I and PD-L1 expression on tumor cells and exhibits anti-tumor activity both as a monotherapy and in combination with PD-1/PD-L1 checkpoint inhibitors.
