Overview

NKTR-422 is a novel, polyethylene glycol (PEG)-modified hematopoietic colony stimulating factor (CSF) protein engineered to selectively modulate the resolution processes of inflammation. It acts by targeting the expansion, reprogramming, and activation of anti-inflammatory tissue resident macrophages. Unlike traditional CSF-1 therapies that require frequent dosing due to rapid clearance, NKTR-422 has a differentiated pharmacokinetic and pharmacodynamic profile with reduced clearance, enabling sustained activity from a single dose. In preclinical models of chronic inflammatory conditions, it demonstrated induction of inflammation resolution and tissue repair markers in tissue resident macrophages without causing monocytosis and enhanced efficacy when combined with standard inflammatory cytokine blockade drugs. The drug is being developed primarily for autoimmune diseases and other acute or chronic inflammatory disorders[1][3][5][7].
