NKX019 + fludarabine + cyclophosphamide is a combination therapy consisting of **NKX019**, an allogeneic, genetically engineered natural killer (NK) cell therapy expressing a chimeric antigen receptor targeting CD19, administered following lymphodepletion with **fludarabine** and **cyclophosphamide**. NKX019 is derived from healthy donor NK cells and utilizes CD3 zeta and OX40 co-stimulatory domains, along with membrane-bound IL-15 for enhanced activation and persistence. Fludarabine and cyclophosphamide are chemotherapeutic agents commonly used for lymphodepletion to facilitate adoptive cell transfer. This regimen is under clinical investigation for relapsed/refractory B-cell malignancies, lupus nephritis, systemic lupus erythematosus, systemic sclerosis, idiopathic inflammatory myopathy, and ANCA-associated vasculitis. The investigational therapy is primarily being developed and trialed by Nkarta.
