Overview

NKX101 is an investigational, allogeneic, off-the-shelf chimeric antigen receptor (CAR) natural killer (NK) cell therapy developed for cancer immunotherapy. It is derived from healthy donor peripheral blood and engineered to express a CAR targeting NKG2D ligands on tumor cells, as well as a membrane-bound form of interleukin-15 (IL15) to enhance persistence and activity. The therapy aims to augment the innate anti-tumor activity of natural killer cells by increasing their ability to recognize and kill malignant cells expressing stress-induced ligands recognized by the NKG2D receptor. Preclinical studies have shown that engineering with the proprietary NKG2D-based CAR significantly increases cytotoxicity compared to non-engineered NK cells. Clinical development has focused primarily on relapsed or refractory acute myeloid leukemia (AML), with additional investigation in other hematologic malignancies and solid tumors[1][3][4][5][6].
