NL variant CD27 CAR-NK is an experimental chimeric antigen receptor (CAR) natural killer (NK) cell therapy optimized via deep learning to target CD70-expressing malignancies, such as acute myeloid leukemia (AML) and renal cell carcinoma (RCC). The therapy utilizes a "natural ligand" (NL) binder derived from the extracellular domain of CD27, the physiological receptor for CD70. Researchers at the University of California, San Francisco (UCSF) and UC Berkeley employed machine learning algorithms to identify specific mutations in the CD27 binder that enhance its affinity for CD70 by approximately two-fold, primarily through a reduced dissociation rate. This optimization results in improved immune synapse stabilization and superior anti-tumor efficacy in preclinical models compared to wildtype CD27-based CARs. The construct is delivered into primary NK cells using gamma-retroviral vectors for potential clinical translation in hematologic and solid tumors.
