NL variant CD27 CAR-T is an investigational chimeric antigen receptor (CAR) T-cell therapy targeting CD70, developed by researchers at the University of California, San Francisco (UCSF) and the University of California, Berkeley. The therapy utilizes a natural ligand (NL) binder derived from the extracellular domain of CD27, the physiological receptor for CD70. Through machine learning-guided optimization, a specific variant of the CD27 ligand was engineered to possess a 2-fold higher affinity for CD70 compared to the wild-type ligand, primarily driven by a slower dissociation rate. This optimization enhances immune synapse stabilization and signaling, resulting in superior anti-tumor efficacy and persistence in preclinical models of acute myeloid leukemia (AML). The platform is also being evaluated in CAR-NK cell formats and for other CD70-expressing malignancies, such as renal cell carcinoma.
