NL019 is a next-generation, dual-target antibody-drug conjugate (ADC) developed by Naliean Therapeutics using its proprietary NanoOne™ platform. It is designed to simultaneously target Trophoblast cell-surface antigen 2 (TROP2) and Nectin cell adhesion molecule 4 (NECTIN4), two antigens widely expressed in epithelial malignancies. The molecule is constructed using modular VHH nanobody fragments, resulting in a smaller molecular size (<54 kDa) compared to traditional IgG-based ADCs, which enhances tumor penetration. NL019 features site-specific dual-payload conjugation of a microtubule inhibitor via a stable, hydrophilic linker. The design excludes the Fc domain to reduce Fc receptor-mediated toxicity while maintaining an optimized pharmacokinetic profile. Preclinical data indicate potent in vitro cytotoxicity and superior in vivo antitumor efficacy in various xenograft models compared to single-target TROP2 or NECTIN4 ADCs.
