Drug intelligence / Profile preview
NLGM1 is a nanoliposomal formulation of the monosialoganglioside GM1 (monosialotetrahexosylganglioside), a glycosphingolipid naturally found in cell membranes. Developed primarily by researchers at the Medical College of Wisconsin, NLGM1 is being investigated for its cytoprotective and vasoprotective properties in conditions characterized by endothelial dysfunction and oxidative stress, such as AL amyloidosis, ischemic stroke, and medin-induced vascular disease. The formulation works by restoring the activity of endothelial nitric oxide synthase (eNOS), thereby increasing nitric oxide production and improving vasoreactivity. Additionally, NLGM1 acts as an antioxidant by reducing the levels of reactive oxygen species (ROS) and peroxynitrite, and by modulating the Nrf-2 stress response pathway. The nanoliposomal carrier enhances the delivery and integration of GM1 into target cell membranes compared to the free form of the ganglioside.
