Drug intelligence / Profile preview
NLGN4X-TCR-T cells are T cell receptor (TCR)-engineered T cells designed to target neuroligin 4 X-linked (NLGN4X), a tumor-associated antigen overexpressed in human gliomas, including glioblastoma. The TCR was discovered via single-cell TCR sequencing of NLGN4X-tetramer-sorted T cells from a glioblastoma patient who had been vaccinated with a multi-peptide vaccine containing the HLA-A*02-restricted NLGN4X131-139 epitope. Transduced into primary human T cells, these TCR-T cells demonstrate antigen-specific cytotoxicity and polyfunctionality against NLGN4X-expressing glioma cells. In preclinical models, intracerebroventricular administration of NLGN4X-TCR-T cells has been shown to prolong survival and induce tumor regression.
