NlpD is a bacterial protein-based inhibitor of RNA Polymerase II being developed by HAMLET BioPharma (formerly HAMLET Pharma) for the treatment of bacterial infections and inflammation. Derived from commensal bacteria, NlpD enters human cells and suppresses the activity of RNA Polymerase II, thereby blocking the expression of disease-related genes. Unlike traditional antibiotics that target bacteria directly, NlpD modulates the host's immune response to prevent over-activation, reduce harmful inflammation, and restore natural antibacterial defenses. The drug is currently in preclinical development, with studies demonstrating potent therapeutic effects in bacterial infection models by targeting the host's disease response rather than the pathogens themselves.
