Drug intelligence / Profile preview
NLRP3-TRAP (Temporally Restricted Assembled inflammasome Platform) is an optogenetic system designed to inhibit the NLRP3 inflammasome in microglia for the treatment of neuropathic pain. Developed by the Institute for Basic Science, the system consists of NLRP3 tethered to a CIBN optogenetic module and co-expressed with a blue-light-responsive CRY2 variant (E281A, A9). Upon blue-light illumination, the system enables spatiotemporally precise trapping and clustering of assembled NLRP3 inflammasomes, thereby attenuating downstream inflammasome signaling, reducing cleaved caspase-1 and mature IL-1β levels, and decreasing microglial activation. For in vivo delivery, NLRP3-TRAP is encapsulated in PLGA nanoparticles, which are preferentially internalized by spinal microglia following intrathecal injection.
