NM-198 is a novel RNA interference (RNAi)-based therapeutic candidate developed by Speratum Biopharma, Inc. It harnesses the tumor-suppressing power of microRNA miR-198 to target multiple oncogenic pathways involved in cancer progression. The drug acts as a mesothelin (MSLN) inhibitor through RNA interference mechanisms, downregulating factors such as Valosin-containing protein (VCP) that are implicated in autophagy and drug resistance in pancreatic ductal adenocarcinoma (PDAC). NM-198 is delivered using Speratum’s proprietary Nano-In™ nanoparticle platform combined with the NoPass™ technology for enhanced targeting specificity and reduced off-target effects. Preclinical studies have shown that NM-198 sensitizes resistant pancreatic cancer models to gemcitabine chemotherapy, significantly reducing tumor growth and metastasis. The therapy aims to overcome multi-drug resistance by disrupting autophagy maturation and other key cancer pathways[1][2][4][5].
