Drug intelligence / Profile preview
NM-PB nanozyme is a biomimetic, hybrid membrane-coated Prussian blue (PB) nanozyme developed for the treatment of ulcerative colitis (UC). It consists of an inorganic Prussian blue core, which possesses potent multi-enzyme mimetic and antioxidant activities, coated with a hybrid cell membrane derived from neutrophils and macrophages. This biomimetic camouflage enhances the nanozyme's biocompatibility, prolongs systemic circulation, and improves targeting efficiency to inflamed colonic lesions. Mechanistically, NM-PB nanozyme scavenges reactive oxygen species (ROS), reduces pro-inflammatory cytokines (such as TNF-alpha, IL-6, and IL-1beta), and promotes the polarization of macrophages from a pro-inflammatory M1 phenotype to an anti-inflammatory M2 phenotype. Furthermore, it activates the Nrf2 signaling pathway, upregulating antioxidant proteins like GPX4 and Slc7a11, which inhibits macrophage ferroptosis and repairs the intestinal mucosal barrier.
