NM100061 is a proprietary topical cream formulation of the tricyclic antidepressant clomipramine, developed by NexMed (later Apricus Biosciences) for the treatment of premature ejaculation (PE). The product utilizes NexMed's proprietary NexACT transdermal delivery technology, which incorporates the permeation enhancer DDAIP HCl (dodecyl 2-(N,N-dimethylamino)propionate) to facilitate the absorption of clomipramine through the penile skin. By delivering the drug topically, NM100061 aims to provide the ejaculatory-delaying benefits of serotonin reuptake inhibition while minimizing the systemic side effects typically associated with oral administration of SSRIs or tricyclic antidepressants. Clinical data from an international pilot study involving 89 patients demonstrated a high satisfaction rate of 84.8% compared to 23.3% for the placebo group, indicating significant efficacy in delaying ejaculation and improving patient satisfaction.
