Overview

NM1332 is a novel small-molecule inhibitor of Interleukin-11 (IL-11) signaling, currently under development by Nyrada for the treatment of arthrofibrosis and other fibrotic conditions. Arthrofibrosis is a debilitating condition characterized by excessive joint scarring and collagen production, often occurring as a complication of osteoarthritis or following surgical trauma such as total knee arthroplasty (TKA). NM1332 targets the IL-11 pathway, which acts as a critical mediator of fibrosis downstream of transforming growth factor-beta 1 (TGF-β1). By inhibiting IL-11 signaling, the drug aims to attenuate the activation of synovial fibroblasts and reduce the overproduction of type I collagen, thereby preventing the stiffness and pain associated with joint fibrosis.
